In this investigation crosses were made between CMS line (IR58025A) possessing “wild abortive” (WA) cytoplasm as female line and five diverse rice varieties/ genotypes as tester (males) and crosses obtained through crossing in a “line X tester” mating design (Kempthorne, 1957). This work was done in kharif season, 2007. For the validation of F1 total genomic DNA was isolated from each F1 plant separately and also from parents by CTAB method and finally SSR was done by two primers RM 258 and RM 247 and a 3% agarose gel was run for the electrophoresis. The molecular analysis on the basis of amplification pattern by RM 258 revealed that crosses IR58025A X Narendra Usar 3, IR58025A X NDRK 5097, IR58025A X 6IRRI, IR58025A X 20 IRRI and IR58025A X N-22 exhibited a DNA band of 155 bp similar to their female parent i.e. IR58025A and the crosses also exhibited 100% spikelet sterility in their bagged panicle revealed that all the male parents were identified as maintainer lines.
But the molecular analysis on the basis of amplification pattern by RM 247 showed that crosses, IR58025A X Narendra Usar 3, IR58025A X NDRK 5097, IR58025A X 6 IRRI, IR58025A X 20 IRRI, IR58025A X N-22 as well as their parents exhibited genome specific DNA band of 155 bp. However, they are similar at nucleotide level or not can only be confirmed after its sequencing.
